BRCA mutations impair DNA repair through homologous recombination. Carriers have higher risks of breast, ovarian, prostate, and pancreatic cancers. Tumors with BRCA mutations are sensitive to platinum chemotherapy and PARP inhibitors (olaparib, rucaparib, niraparib). Testing can be germline (inherited) or somatic (tumor-only).
